You will need a USB cable with a Standard-A connector and a Mini-B connector to access the Memory Stick Duo in your PSP.  Plug the Mini-B side into the top of your PSP, and the Standard-A side into your PC.  Once you have connected these, go into your PSP system software, navigate to the top of the settings list, and select "USB Connection".  Your computer should detect your PSP as a new "Removable Disk Drive", much like a digital camera, which you will be able to access.

You do not need wireless access in order to play against other gamers with your PSP.  You can connect up to 16 PSP systems in the vicinity directly through the ad-hoc mode for without requiring any additional hardware.
